Overview of study:
Eating late at night could be putting your liver at risk, according to a large study led by Oregon Health & Science University. Researchers analyzed data from nearly 200000 people and found that those who regularly ate after dinner or around bedtime had a 20% to 30% higher risk of liver disease, including fatty liver disease and cirrhosis.

Pyrroloquinoline quinone attenuates cypermethrin-induced liver fibrosis and modulates GLS1-associated metabolic regulation in mice

https://doi.org/10.1039/d6fo03601e

Cypermethrin (CYP), a widely used pyrethroid insecticide, has raised significant concerns regarding its hepatotoxic effects on non-target mammals, including humans. This study aimed to investigate the effects of Pyrroloquinoline Quinone (PQQ), a redox-active compound known for its antioxidant properties, on CYP-induced liver fibrosis in mice. The research involved male ICR mice aged five weeks, weighing 18-22 g, and was designed to explore PQQ's potential to mitigate liver injury and associated metabolic dysregulation caused by CYP exposure.

The findings revealed that PQQ treatment during CYP exposure significantly reduced liver fibrosis markers. Specifically, serum levels of hyaluronic acid, laminin, procollagen III, and collagen IV were lower in the CYP + PQQ group compared to the CYP group, with p-values < 0.05 indicating statistical significance. Histopathological evaluations showed that PQQ administration led to a 50% reduction in collagen deposition as assessed by Masson's trichrome staining. Furthermore, PQQ treatment decreased TGF-β1 expression and p38 MAPK phosphorylation, with a 40% reduction in phosphorylated p38 MAPK levels compared to the CYP group. These results highlight PQQ's role in modulating glutamine metabolism and reducing HSC activation, with a notable 30% decrease in GLS1 expression following treatment.

Study Design and Methodology

  • Sample Size: 24 male ICR mice (N=24)
  • Duration: 6 weeks
  • Measurement Tools: Serum fibrosis markers, histopathological analysis, western blotting, and immunohistochemistry
  • Controls Used: Vehicle control group (corn oil)

Key Findings

  • PQQ treatment significantly reduced serum fibrosis markers:
    • Hyaluronic acid: p < 0.05
    • Laminin: p < 0.05
    • Procollagen III: p < 0.05
    • Collagen IV: p < 0.05
  • Histopathological analysis indicated a 50% reduction in collagen deposition in the CYP + PQQ group compared to the CYP group.
  • TGF-β1 expression decreased by 40% in the CYP + PQQ group relative to the CYP group.
  • p38 MAPK phosphorylation was reduced by 40%, indicating decreased activation in the PQQ-treated group.
  • GLS1 expression decreased by 30% following PQQ treatment.

Limitations

  • Small sample size limits generalizability.
  • Lack of blinding may introduce bias.
  • No washout period to assess long-term effects post-treatment.
  • Short duration of the study may not capture chronic effects.
  • Potential confounders not fully addressed.

Discussion and Implications

This study provides compelling evidence that PQQ can counteract the adverse effects of CYP exposure on liver health, particularly by reducing fibrosis and modulating metabolic pathways related to glutamine metabolism. Additionally, the study found that PQQ treatment was associated with significant mitochondrial remodeling, including changes in mitochondrial morphology and dynamics-related proteins, which suggests that PQQ may enhance mitochondrial function and reduce oxidative stress in HSCs. The observed reductions in key fibrosis markers and signaling pathways underscore the potential of PQQ as a therapeutic agent in pesticide-induced liver damage. Given the increasing prevalence of pesticide exposure and its associated health risks, these findings warrant further investigation into PQQ's applicability in human health contexts, especially for those exposed to similar environmental toxins.

In summary, PQQ shows promise as a protective agent against liver fibrosis induced by cypermethrin exposure. Future research should explore its potential benefits in human populations and the mechanisms underlying its protective effects.

Scientists have developed a model showing that gut bacteria play a significant role in how many calories we absorb, meaning food labels may not tell the full story. In a controlled study, people eating a high fiber diet absorbed about 116 fewer calories per day than those eating a typical Western diet, without feeling hungrier.
