Hello all,
I am a biologist biotechnologist with both degrees. I have focused alot on microbiology and cell culture.
I am fascinated with slime molds and true fungi. And I have edited several fungi and protists to produce new colors, make cellulase variants, be bioluminescent ect.
Now ive tried the major and og methods of editing microbes with
Physarum Polycephalum or yellow slime mold.
And tried with tubnifera magna (but this is couldnt find a media to keep it alive, was younger less experienced. still upset as ive looked for another sample for years and never found it.)
But the slime mold never holds onto the new genes or they don't incorporate properly. Ive gotten Physarum Polycephalum to hold onto a plasmid used to make bacteria glow in bio classes. but within a day or 3. it is completely purged.
Now I have been thinking that perhaps I can get it to hold onto plasmids through a different brute force.
I am going to search for some more geenticly acceptable genes for a slime mold as i imagine the bacterial genes don't translate perfectly.
but in the mean time. I have recently forced a culture of ecoli, and 2 cultures of as of yet unidentified bacteria that I found in a contaminated slime mold plate and which I watched my Nesse eat under microscope.
so now I have 3 bacteria that nesee will eat. and all 3 have been forced to take in multiple copies of bioluminescent plasmids.
and now im going to be feeding nesse these cultures daily to see if over time the slime mold will start recognizing the plasmids as just another piece of dna in its single cell beauty.
not super hopeful. but sometimes you just need time. and if nesse doesn't start producing waves of bioluminescs as it pulses after a month. i will give up.
Until I get my next idea.
anyone got any advice?

I was photographing this raspberry slime mold and in the photo I noticed this tiny white dot on a stem. It resembles Comatricha nigra young fruiting body, but it is much smaller than 1 mm.
