Success story Possible biotin recycling fault (biotinidase deficiency) as underlying problem, with great effect on metabolic issues and candida/SIBO

Candida/SIBO most probably have a multitude of causes, but as a person who have tried absolutely everything (and have at times been bed-ridden and almost reached insanity in the valley between hope and despair; MADRS: 20-40), biotin in high doses has been nothing but revolutionary. Not just because of its possible interactions with candida itself - it is assumed to have dehyfinating effects - but because of metabolic and mucosal health reasons.

I have never been able to tolerate fasting well, and I currently blame that on biotin deficiency, since it is crucial for gluconeogenesis and possibly causes CoA-sequestration that disrupts fatty acid metabolism. As a kid I fainted several times because of what I now think was hypoglycemic episodes, and as an adult I have tried fasting up to 60 hours, without having a single ketone in my blood or urine, and feeling utmost shit. 5 mg of biotin gets me into ketosis within 3 hours, given that I am in a fasted state. This might be mediated through many pathways, but possible ones are the mentioned CoA sequastration, or the high amounts of succinyl-coa needed to convert the ketones AcAc into BHB (I will spare the rest of the details, because I don't have them right now, hehe). I would love some suggestions for why biotin deficiency would hinder ketosis, as everything I have found says the opposite would happen. Namely that ketones should go drastically up when acetyl-CoA loses its biotin-dependent oxaloacetate to enter the TCA-cycle with. Anyways, my experiments contradicts this. And also, since 5 mg of biotin only gets me into ketosis 24-36 hours before it tapers off to eventually zero again, tells me that there is something wrong with recycling, not mere uptake. I also think the malonyl-CoA pathway, which creates fatty acids essential to barrier function, that biotin engages, is of great importance in allowing candida to thrive.

Biotinidase is responisble for this recycling, and if one lacks the enzyme, this will reduce the amount of biotin available. Biotinidase deficiency in its most harrowing form causes a less than 10% effect of the enzyme, but less extreme cases, partial deficiencies, said to still be rare, range from 10-30% enzyme function. Chris Masterjohn (nutritionist) nevertheless posits that about 1/30 people could benefit from high doses, so the partial deficiency pool might be broader and more frequent than assumed. These kinds of deficiencies is said to not normally present symptoms before one has a grave infection or reaches adulthood. Where the former is exactly what happened to me. But no doctor/practioner, in my country (Scandinavic) at least, tests for these deficiencies, and the bio-markers it gives are vague or non-existent, in my experience. Also, I am born way before they started to check new-borns for it in 2012.

Since getting an infection more than 10 years ago (food poisoning that really messed with my gut), in my late teens, I have never been quite the same, and my stomach has just gotten worse each year. The light in me slowly perished; I thought I was depressed, that I was living out of touch with myself or whatever. I thought existentialism would fix it, or sleep, meditation, or working out, light therapy, etc etc, behaviour behaviour behaviour. After some years of diminishing health, I started nutritionally optimalizing all that was in my control; from thyroid stuff, methylation, vitamin A through K, minerals, proteins, fats, herbs, anti-biotics, anything really. Which variably helped, but I was still brain fogged, with mild to moderate candida infections on my tongue, thighs, genitals, peri-anal area, etc., smelling funny under my arms, hair oily as hell, stomach most of the time feeling bad, full, nervous, etc., feeling overall depressed and exhausted, could not read a book if I tried (this is perhaps the saddest thing, cannot even watch a movie as distraction). Sex life was (have not tried in a while, so might still be, for other reasons hehe) horrible, and filled with shame due to lack of physical sensation, or if anything pain, with a mucosa as dry as Death valley. Prescribed topical anti-fungals would work for the duration I used them, before coming back a couple days later. Sugar was never a part of my diet (although I craved carbs such as popcorn, potato chips), have never been overweight (rather always been the lanky type), and do live quite thruthfully, to be frank:-)

My thought was, still is, that there are no good reasons that a seemingly healthy body and immune system should have candida indefinitely, especially after anti-fungal cures (MCT-oil, garlic, black seed oil, oregano oil etc.). And it is somewhat absurd to think that one has to take anti-fungals daily to keep it in check - there must be something underlying, a blockade. This is a point somewhat tangible to Chris Masterjohn's idea that energy metabolism governs everything - that is, fix your own resources, and the body will recover itself (anti-fungals might be needed to catalyze it, or not). Anyways, having a low energy output that causes 10-30% cognitive/physical endurance function surely must reflect somewhat in the gut as well (in addition to the fatty acid component of biotin mentioned). After having tested several times to get into ketosis with many different tactics, and also, prior to that, randomly experimenting with biotin from time to time (hypothesized an oxalate issue, and as I said, I tried everything just to be sure), I suddenly went from zero ketones after 27 hours of fasting, to "low" (which is decent) amounts on the urine stick within the next three hours - confirmed by me feeling a lot better. The benefits from experimenting with biotin was not clear to me when I was filled with carbohydrates, as I judged the die off symptoms as something that was wrong, or as a transient oxalate dumping (see Chris Masterjohn's substack for that connection).

The results have not disappointed, and I am of course terrified of falling back into it - 10 years of emptiness is enough. I still probably have a couple of weeks until the rest of the fog vanishes, and a month or two to go fixing my gut, but I have nonetheless six full days with 1.5-3.0 levels of ketones in my blood behind me, and a gradual, but significant increase in mood, energy and skin symptoms (mucosal skin is drastically better). I would never have been able to write this in that former exhausted, terrifying state - I actually felt like a nine year old kid, cognitively. And I maintain that the die offs are also directly caused by the timing of the dose, so if you don't have a biotin recycling problem, it can still be of value to try for the possible candida-weakening effect (assuming that die offs actually are a positive sign. I do, hehe). The assumed die offs have been ghoulish, but surely decreasing each day. I find that molybdenum, vitamin C and vitamin A helps the most in managing symptoms.

Yesterday I cried of joy because I was feeling again. That is real, unexpected internal movements that seems almost electric and spontanous. I have found myself ruminating on problems or thinking of other people's gestures again, laughing about how I would usually deem that as something negative, but now as mere potentials I could canalize or regulate! Oh, and that sensation of managing intensities, increasing or decreasing my enthusiasms, like ripples in a pond, where I am the one holding the rocks. I had forgotten it existed. All this is to say, I/we am/was/were/are not brain damaged! And that there are still layers beneath the sheath.

I am not saying this is your case: MMC, immune-function, anti-fungals and other deficiencies might be the solution or reason for your pathology. For me, all of those supporting and killing mechanisms have helped. But if you have had some similar metabolic problems, or not for that matter, then a week, or a month, or indefinent use of 5 mg biotin has no known toxicity risks. Chris Masterjohn speaks anectdotally of him getting clumsy after a couple of weeks of high doses, and some small increase in lactate, so that is that. Just remember to take it 3-4 hours away from your B5-intake, as they share the SMVT transporter.

For the ones I have sparked some hope in, but to your despair it does not or have not worked - I am terribly sorry. I feel ambigous about these exclamations of betterment, and self-aware about the lack of control I have, better yet the road to go - I should speak conservatively. What has been the most help for me is this community, posts like these, where I have seen other people struggling or finding betterment. In my view, do not give up the hope, you have the right to feel okay, even though people think you are trying to optimize. That goes to all of you that are struggling, even myself, who feel totally lost: it feels like battling something as bad as cancer imo - it just does not kill you fast enough. There are very few places were one can find validation for this, and I would say the condition has this double whammy of loneliness; it does not make sense to most people, and you therefore have to hold onto it on your own. But then again, "you" are not even there for yourself.

I wish you all the best!

Tl;dr: Biotin in doses of 5 mg have solved my fasting intolerance and metabolic issues, and seems to rapidly have changed my gut, mood and skin issues for the better in six days. The assumed direct anti-candida effect is dose-respondent (100 mg gives more die off than 5 mg) and possibly mediated through anti-hyphinating effects, in addition to fixing metabolic issues and possibly barrier function.

I'll update in a month or so, and try to answer if anything is unclear.

Should not need to say, but only carbon-based flesh has been used to create this text.

Symptoms CONSTANT YEAST AKA CANDIDA AND MUCUS IN STOOL, ETC? I've learned that it's a parasite. That's why it's moving, etc. It's definitely a parasite that causes more yeast, for your gut to be off balance. THE PARASITE CAUSES THE INBALANCE IN THE GUT! Spoiler

It's a parasite. Mines is at the state where it's moving..... they are moving. It's definitely a parasite. Nothing will kill this but imma go back to the old school....lil bl3@ch and water solution.....not too much bl3@ch but it's the only thing that will do it. I'll get a tub of water and add a top or two full of bl3@ch and do rinses. Im going to bathe in it, etc. IT'S A PARASITE GUYS! THEY'RE PARASITES! NOBODY IS DRINKING BLEACH....DO Y'ALL EVER THINK? I've been to many doctors...even parasite doctor and given them stools and guess what they said? "It's all in your head and you're just paranoid!"

Any other ways to kill a smart parasite that's in your vagina and anal region and has enough intelligence to crawl outside of the body when they know medicine is inside and crawl back in when it's safe? I thought not. It's linked to it....IT'S THE REASONING BEHIND IT. Y'all have to start reading. Again, the parasite is what is causing the constant yeast infections. Y'all are so far lost. I've dealt with this for years and I've killed it at least twice before. I KNOW WTF IM TALKING ABOUT. IT'S STAGES TO IT. BUT Y'ALL KEEP GOING OFF WHAT THE DOCTORS TELL Y'ALL BUT THE SAME MFS Y'ALL ARE LSITENING TO CAN'T AND HASN'T HEALED Y'ALL YET.

Symptoms Has my candida come back?

I was having really bad constipation and bloating symptoms and went to a naturopath. One of the tests was for candida and I ended up having it and was taking Candida Bactin AR for about 3 months. I've stopped taking the pills about 2 months ago but I've recently noticed that I'm experiencing symptoms such as being itchy and having bad BO that i experienced when I was having die off symptoms.

I'm wondering if its a possibility that it never fully died off and the candida is still in my system or if its something else?

Symptoms Positive for Candida

Sometime ago, I believe last year, I went to the doctor and tested for Candida.

I was confused by this because…the doctor said she’d prescribe me Candida. Which meant my yeast infection was still present, but I didn’t feel like it was lol.

I just now realized that yeast infection and Candida overgrowth are not the same. I am going through an overgrowth after my period for some reason and it got increasingly worse. My nether regions went dry and I panicked. I was trying to figure out what was going on. It didn’t feel like a yeast infection , but something was off. No itchiness, discharge , or smells (early on it did smell kind of yeast like).

My butt was itchy, my head was itchy, my ears are itchy, I was gassy, I had acne and so many brain farts. I just took fluconazole and feel a little better.

Honestly, I have no problem quitting sugar or whatever is needed. It’s just the side effects. I’ve never had these before, and I’ve taking fluconazole with no issue. My head is tingly, my stomach gurgled, and TMI! Yeast came out my vagina. wtf???? It’s like it just got pushed out after I took it.

I’ve quickly cut a lot of things out of my diet. Hopefully this goes away soon. It seems like every-time I eat sugar or change my routine it, I get a yeast infection or something.

Supplements Chronic Vaginal Yeast Infections

I’ve been having recurrent yeast infections for 6 months straight. Ive only ever had 2 in my life before this, but after having period sex I cannot get this one away. (not sure if that could be adding to the reason of recurring)
Note: I haven’t had sex at all in these 6 months

Things i’ve tried
- 6 months of fluconazole treatment (currently on)

- Tested for mycoplasma & ureaplasma
(negative for these and all stds/stis)

- oil of oregano

- sensitive dove soap, clean & clear laundry detergent, cotton underwear only and no underwear at night

- pure cranberry juice + kefir & kimchi

- candida diet

- Probiotics daily

I’m currently thinking of trying NAC supplements but if anyone has anything else or comments please help i cannot deal with this for the rest of my life

Personal anecdote Any advice for improvement?

I'm going through something quite annoying like balanitis, for almost 6 months during a shower after soaping my penis I had a rare burning and when cleaning with water I noticed two small red spots on the glans, I didn't really care because I thought it was the soap (which was neutral antibacterial) that had caused me the irritation and redness, and it regularly goes away on its own, but the problem continued, which still 1 month later I suffered from a strong urinary and kidney infection because the doctor prescribed me the use of antibiotics and medication (LEVOFLOXACINO, phenazopyridine and composite hyoscin), as well as medical studies, however I suppose that with the use of the strong antibiotic the problem worsened and practically spread throughout the glans except for the part of the urethra and frenulum, forming almost a ring, and that's when symptoms such as itching, burning with sensitivity, rare smell and slight somewhat creamy white discharge began, this happened in a span of time After 2 months of getting better because the doctor ordered me a different diet, lactobacilli, among others..., after those 2 months of improvement due to infection I prescribe miconazole cream almost 20 days and the truth is that I did not improve after a short pause and I prescribe Ketoconazole cream for almost 15 days and because there was no improvement, so after 1 week of pause I prescribe barmicil cream for 5 days and simultaneous fluconazole for 15 days, the steroid cream improved inflammation and redness but it did not improve either. After days I return in the same way and the fluconazole relieves the itching 90% and the discharge stopped, but I don't know if this will come back, I'm actually a little tired of this and I don't know if it would get better soon, I have an appointment scheduled soon with the doctor, sorry if the text was very extensive but I wanted to give a broad context and with this I want to know if someone has been through something similar and how I manage to solve this problem, likewise their tips or tips to improve, (by the way, I want to know if someone has been through something similar and how I can solve this problem,). My hygiene that I maintain is: daily bath, I use neutral soap and I try to dry the used area well (paper and air, every time I urinate because I still clean the possible traces of urine, likewise I am not circumcised but I can retract the foreskin well without problems to clean the area). Does anyone have any advice for improvement?

Symptoms Is it normal for calprotectin levels to rise to 245 a month after completely eliminating *Candida*?

I’ve been *Candida*-free for a month and am strictly following the diet while taking turmeric, quercetin, and vitamin C to lower accumulated histamine. During this time, I developed herpes and mild tracheobronchitis, which caused dizziness to return and sent my histamine levels and depression skyrocketing. I’m taking 0.5 mg of clonazepam to sleep, and I woke up feeling great today, but is that number normal? Shouldn't it have gone down by now? I admit that in August I started eating flour and dairy; I think that was the main reason the levels spiked. For the past four days, I’ve been treating it with 2g of mesalazine (Pentasa) every 24 hours, and I’m noticing the dizziness gradually subsiding.

General Discussion Has anyone used biofilm distruptors in the treatment for Candida?

Hello dears,

I have this Candida overgrowth that showed up in a dysbiosis test, along with E-coli and Citobacter as well overpopullated. Fecal ph 7,5. Good bacteria down.
I had 2 treatments in 4 months that failed me. They were given by doctors.
First one was with fluconazole and Megasporebiotic, that reduced the Candida but not within the normal range. Still big overgrowth. The thing is, Ecoli was doubled during this treatment so we needed to get a second round.
Second round was with nystatin and rifaximin, 14 days. After this one Candida was not moving down at all, ecoli was going half but still overpopulated and Citobacter exploded. My intestines were literaly burning inside.
The alergies from food were strait up from hell. Histamine intolerance at its peak.
I needed 1 month to use L-glutamine, Sodium butirate, MegaIg2000 and boulardi to calm down my inside burning.
I lost during this journey 10 kg. I am below limit, for my height. And weak as hell.
So i am asking this, for when I manage to get a bit stronger and close my mucosa linning, Has anyone tryed Biofilm distruptors?
I was not given these as part of the treatment. I suspect this might be the cause why I failed every time.
Please advise.

Success story How my candida overgrowth was identified and fixed

Like some before me I have created a Reddit account for this post, to share my experience with yeast overgrowth and try to help others as I have been helped on here! This is going to be a bit long!

My problems started around 2 years ago when my beloved dog was diagnosed with heart cancer, and died a while later. The stress and grief were intense and when he died I lost my appetite. I developed wicked exhaustion and hair loss, both thinning and in patches. Bloodwork showed that my ferritin was very low and my thyroid function was wonky (I was previously diagnosed with hypothyroidism). Taking iron supplements and increasing Synthroid didn’t help, and I was eventually given ferritin in an IV. But shortly after levels of ferritin would start free falling as my body wasn’t absorbing it. I then started having major stomach pain when eating high-fat foods or drinking wine, carbonated water, etc. I would wake up in the night feeling nauseous fairly often. I was tested for gallstones but didn’t have them. My GP would test things when I asked but was unbothered by what could be causing all these issues and was not interested in finding out, so I found a functional medicine doctor.

I paid for 3 tests (I’m in Canada so this sucked in a new way) and they came back diagnosing me with a raging yeast overgrowth (12x the healthy upper limit), bacterial overgrowth and resulting leaky gut. I had no idea there was a candida overgrowth! The most helpful test was the Organic Acids Test (OAT), test was a urine test that measures metabolic byproducts of microbial activity. It identified aspergillus was causing the yeast/fungal overgrowth and flagged a marker produced by Clostridia bacteria as well. I started Candida STOP that day, mostly avoided added sugar (but nothing ínstense and I definitely had ice cream a few times) in my diet and cut out alcohol. It was a brutal 6 weeks of bloating and pain but it worked and my stomach pH has gone from 3.5 to 5.5 in 2 months! The insane bloating and pain is gone, and with tonnes of other supplements my gut barrier is rebounding (note: a GI360 test identified the gut disbyosis).

But HERE IS MY KEY POINT! This test identified the exact yeast and bacterium causing my problems. If I had been given fluconazole by my GP, it would have done nothing because aspergillus does not respond to it! So it was so important to find out exactly which yeast/bacterium was in there. I was also told that just cutting sugar is insufficient because the yeast will eventually start eating anything they can get.

It also turned out that aspergillus are ‘iron scavengers’ so they were consuming any iron that wasn’t leaking out of my weak gut barrier. And aspergillus produces tartaric acid, which was disrupting my metabolism and contributing to very low energy levels. All my symptoms were a result of this overgrowth triggered by stress, but the symptoms didn’t point to candida at first.

Anyway my hair is growing back, my thyroid is starting to even out and I can drink wine again! I wanted to share, I hope this helps  :)

Success story Finally I understood it’s not infection nor Candida

Hello everyone I hope you’re all doing well, first of all I would like to thank everyone tried to help in this community specially my dear friends : Abominable_phoenix and Dr. Eric

Second, I understood that my problem with some fruits, nuts, chocolate, coffee and tea was caused by something called tannins intolerance plus vitamins and minerals deficiency.

Finally I would like to give small advice to those who’s trying to restore their gut bacteria the best thing I’ve tried was Sauerkraut. Good luck to you all and I hope that you can find the main cause as I did.

PS: this thing started with me after I had long Covid.

Symptoms Has anyone successfully treated fluconazole-resistant Candida albicans with nystatin vaginal suppositories? 14 vs. 21 days?

Thumbnail
1 Upvotes

I’ve been dealing with recurrent, fluconazole-resistant Candida albicans for years, and I’m currently trying nystatin vaginal suppositories (100,000 units nightly).
Tonight will be my 14th and final night of treatment. I’ve definitely noticed improvement in my symptoms, especially the itching, but I think I’m still seeing some yeast/discharge, so I’m worried the infection hasn’t completely cleared.
Here’s where I’m confused:
My infectious disease doctor recommended completing 21 days of nystatin, but my gynecologist recommended 14 days, explaining that if the medication was going to work, the infection should have cleared within those 14 days.
I’m hesitant to stop at 14 days when I’m finally experiencing some relief, especially since I’ve struggled with resistant yeast infections for so long. At the same time, I don’t know if extending treatment to 21 days would actually make a difference.
Has anyone here had success treating resistant Candida albicans with nystatin vaginal suppositories?
● Did you use them for 14 or 21 days?
● Were you still experiencing symptoms or seeing yeast toward the end of treatment?
● Did extending treatment help clear the infection?
● Did your infection eventually come back, or have you remained yeast-free?
I’d really appreciate hearing about anyone’s experience, especially if you’ve dealt with azole-resistant Candida albicans.

Diet Has anyone managed to heal from Candida just by cutting refined sugar alone?

I have recurring Candida issues but also admit I have a pretty wild sugar addiction. I can get off it for days but eventually I feel like I’m going insane and I’ll binge.

This is really affecting my health and now I’m on an immunosuppressant for eczema which is probably Candida related… and now I’m getting chronic sinus infections.

I have a small child and have recently gone back to work. I am finding it really overwhelming doing an entire diet change and I know how much commitment that needs.

Has anyone managed to rid themselves of Candida / their main symptoms by just quitting refined sugar?

I struggle eating proper meals but these are important to stop me crashing and needing sugar. I need something easy during the day whilst working from home and I’d like to fall back on something easy and comforting like a toastie and some soup etc.

Symptoms Candida Glabrata. Anyone who was able to eradicate it how? & what were your symptoms? Any burning up feeling in your pelvis groin area. && even caused you a flank pain and pelvic pain, fungal UTIs? Bc same.

For 2 years at least I have been dealing with this chronic yeast infection but it wasn’t until last month that I became very ill.

I feel like this yeast is colonizing me.
And activating my immune system so bad that I have chronic inflammation and bad anxiety. It even caused me a uti which makes sense why I have had shivers for a whole month. On and off pelvic and flank pain. Discharge.

There was a point that it I felt like oral thrush. Ear aches. Sore throat.

My belly button had yeast! Crusting and foul smelling with watery discharge and red inside.

My hair has seb derm.

This can’t be normal.

Anyone else with chronic uti symptoms but most of 99.9% of the time it’s not a uti. This is the first time they actually say it was a fungal uti and yeast infection as well.

Supplements Reaction to Saccharomyces Boulardii

Has anyone else had a bad reaction to Saccharomyces Boulardii?

I just finished a month long candida diet where I was taking lots of antifungal herbs along with a Biofilm disruptor. I think they helped clear out a lot of the yeast but also probably irritated my gut lining, because now I’m noticing sensitivities to things I never had issues with before (like low acid coffee).

This morning I tried taking one capsule of Now S. Boulardii on an empty stomach, and an hour later I was overcome with the worst brain fog of my life, like a hangover after drinking 15 beers the night before.

Could this be caused by the supplement? I’ve always heard it’s super beneficial, but is it possible that my inflamed gut is so sensitive to yeast that any type at all gives me extreme brain fog?

I also had this reaction of extreme brain fog/drowsiness a couple times when I slipped up on the diet and ate more carbs than I should have.

Would love to hear if anyone else has had a reaction like this!
